The primary goal of the thesis was to test the eDNA qPCR protocol. For this
are the Enns and Johnsbach in the area of the Gesäuse National Park for sampling
have been selected. It has been tested whether graylings (Thymallus thymallus) rise into the Johnsbach or not.
The eDNA sampling has five times at 3 times between the end of April and the end of May 2019.
Positions of the Enns and in 2 places of the Johnsbach took place. It’s at every rehearsal site
a total of 1 litre of water has been pressed through filter to collect the eDNA in it. The
Samples were then extracted in the laboratory and tested on the basis of an qPCR, whether therein
Ashes of DNA.
The protocol has worked in both the Enns and Johnsbach, since at any point
at least one replica amplified. It has therefore been found that the Protocol also
in larger rivers such as the Enns, in relation to the size of the river the ashes
Population density is low, works. Also in Johnsbach, where no grayling awaits
few replicas have been amplified, and it can be assumed that ashes
at least ascend to the spot of pale stone.
